Organ bath technique

 

Post-thaw functional changes are 

investigated in organ bath experiments.

 

Tissue preparation

Tissue samples are placed into RPMI culture 

medium and transported to the laboratory to 

be cleaned of loose connective tissue, cut into 

rings of about 2.5 mm length and mounted

between two L-shaped hooks of stainless steel 

wire.

 

 

 

 

In vitro experiments

Vasular rings are suspended in 10 ml organ baths containing Krebs-Henseleit solution at 37°C, gassed continuously with 5% CO2 in oxygen. Changes in tone are recorded isometrically under a resting tension of 1g with electromechanical transducers and a potentiometric recorder.
